چکیده

There have been many investigations of the oxidation-reduction potential of the cytochrome c system. Some of the results are concordant, though none at the time this work was undertaken included a complete potentiometric titration of the system over a range of pH values. Potentials have been reported for the system at varying pH values, usually within a narrow range bordering on the physiological. These results are not concordant, the most divergent being those reported by Green (10). The results of previous workers are summarized in Table I. We report here complete potentiometric titrations, both oxidative and reductive, of the cytochrome c system at different pH and E’,, values for the system throughout the pH range, 0.4 to 10.0. Since undertaking this work, a report of Paul (16) has appeared which covers the range of pH from 4 to 8.0 but with results and conclusions somewhat different from ours.

Site-directed mutagenesis of tetraheme cytochrome c3. Modification of oxidoreduction potentials after heme axial ligand replacement.

The nature of the axial ligands of a heme group is an important factor in maintaining the oxidation-reduction potential of a c-type cytochrome. Cytochrome c3 from Desulfovibrio vulgaris Hildenborough contains four bis-histidinyl coordinated hemes with low oxidation-reduction potentials. Site-directed mutagenesis was used to generate a mutant in which histidine 70, the sixth axial ligand of heme...

Reaction of Ascorbic Acid with Cytochrome

Rate constants for reduction of cytochrome bsel by internal ascorbate (kOA) and oxidation by external ferricyanide (klF) were determined as a function of pH from rates of steady-state electron transfer across chromaffin-vesicle membranes. The pH dependence of electron transfer from cytochrome b56l to ferricyanide (klF) may be attributed to the pH dependence of the membrane surface potential. Th...

Superoxide anion generation by the cytochrome bc1 complex.

We have measured the rates of superoxide anion generation by cytochrome bc(1) complexes isolated from bovine heart and yeast mitochondria and by cytochrome bc(1) complexes from yeast mutants in which the midpoint potentials of the cytochrome b hemes and the Rieske iron-sulfur cluster were altered by mutations in those proteins. With all of the bc(1) complexes the rate of superoxide anion produc...

CO-binding c-type cytochromes and a high-potential cytochrome c in Nitrosomonas europaea.

The purification of two soluble CO-binding cytochromes c from Nitrosomonas europaea is described. Cytochrome cCO-550 ran on sodium dodecyl sulphate/polyacrylamide-gel electrophoresis with an apparent Mr of 32 000, whereas for cytochrome cCO-552 the apparent Mr was 16 000. Redox potentials (Em, 7) were determined as +140 and -50mV respectively. Cytochrome cCO-550 was co-purified with a cytochrom...

The Respiratory Chain of Plant Mitochondria: IX. Oxidation-Reduction Potentials of the Cytochromes of Mung Bean Mitochondria.

The oxidation-reduction potentials of the cytochromes of the respiratory chain of mung bean (Phaseolus aureus) mitochondria have been measured under strictly anaerobic conditions with a combined spectrophotometric/potentiometric method. The midpoint potentials at pH 7.2 are as follows: cytochrome a: +190 millivolts; a(3): +380 millivolts; b(553): +75 millivolts; b(557): +42 millivolts; b(562): ...
